Futhermore, there are no animal models on renal IR injury during PN. This study aims to develop an optimal animal model for renal IR injury that occurs during PN and analyze the efficacy and safety of phosphodiesterase type 5 inhibitor (PDE5i)'s renal protective function. METHODS: Animal model of renal IR injury was established via PN and oral administration of daily tadalafil (5 mg/kg) to 6-week-old male mice for 2 weeks before PN. The degree of functional, histological, and molecular injury was analyzed through blood and kidney tissue. RESULTS: There was no significant difference in serum creatinine and the glomerular filtration rate between tadalafil group and control group. However, histologically, the number of glomerulus increased significantly in tadalafil group. Massons' Trichrome staining confirmed that fibrosis, which increased in the renal IR group, was partially suppressed in the tadalafil group. Immunologic analysis showed that expression of TNF-α, IL-1β, IL-6, iNOS, and PDE5 reduced significantly in tadalafil group. Molecular biological analysis showed that expression of kidney MPO reduced significantly in tadalafil group. Immunohistochemistry (IHC) confirmed that T cells, which were increased in the renal IR injury group, were partially reduced in the tadalafil group through CD4 (T helper cell) and CD8 (cytotoxic T cell) makers. And also IHC showed that apoptosis and inflammatory responses through p53 and eNOS were partially reduced in the tadalafil group. CONCLUSIONS: We developed optimal animal model of renal IR injury during PN and verified renal protective effect of PDE5i.
